Early dopaminergic loss reshaped corticostriatal synaptic gene networks in a region-specific way: PFC modules were enriched for trans-synaptic signalling genes, while STR changes centred on excitatory neuronal signatures and extracellular matrix genes.
The darkred module was associated with trans-synaptic signaling and chemical synaptic transmission and was preferentially associated with the PFC. STR DEGs were enriched for striatal development, locomotion, and extracellular matrix-related processes, and were predominantly enriched in excitatory neuronal signatures.
